    Exploring Agricultural Production Systems and Their Fundamental Components with System Dynamics Modelling

    Agricultural production in the United States is undergoing marked changes due to rapid shifts in consumer demands, input costs, and concerns for food safety and environmental impact. Agricultural production systems are comprised of multidimensional components and drivers that interact in complex ways to influence production sustainability. In a mixed-methods approach, we combine qualitative and quantitative data to develop and simulate a system dynamics model that explores the systemic interaction of these drivers on the economic, environmental and social sustainability of agricultural production. We then use this model to evaluate the role of each driver in determining the differences in sustainability between three distinct production systems: crops only, livestock only, and an integrated crops and livestock system. The result from these modelling efforts found that the greatest potential for sustainability existed with the crops only production system. While this study presents a stand-alone contribution to sector knowledge and practice, it encourages future research in this sector that employs similar systems-based methods to enable more sustainable practices and policies within agricultural production

    Agrarian Myth: Its History and Use by the Australian Country Party

    It is widely acknowledged that agrarian myth was an important component of the Australian Country Party's ideology. This thesis undertakes a detailed examination of the nature of Australian agrarian myth and the manner of its use by the Country Party. The first chapter provides a theoretical framework for the study of myth and its relationship with ideology. The focus of the work then shifts to an examination of the characteristics of agrarian myth, first in Europe and then in America, where the land and the circumstances of its settlement brought about changes in the imported mythology. Some discussion of myth in the European tradition is essential, first to illustrate the strength and duration of the myth which Australia has inherited, and second to provide a model with which Australian agrarian myth can be compared. This also provides the reason for considering rural myth in the American context

    The Borders and Frontiers of Salman Rushdie's 'Literary Project': Perspectives of Identity, Culture and Celebrity Drawn from 'Fury' and 'Step Across This Line'

    Salman Rushdie claims borders and frontiers are "at the heart of [his] literary project" . Given the diverse tone, thematic range and structural diversity of his literature, how are borders and frontiers manifest throughout his project? Compounding this question, how are we to intellectually engage borders and frontiers both within and beyond the context of Rushdie's writing? The term border may infer limits, restrictions, clarity and delineation, yet borders can also be records of transition, transgression, conflict and challenge. Similarly, what are we to make of the spaces in-between, those ambiguous regions of the frontier that amorphously envelop and recede away from borders? This thesis seeks to qualify Rushdie's premise through a concerted examination of five vital facets of borders and frontiers. Firstly, conceptual accounts of borders and frontiers are engaged in a manner designed to delineate real and imagined, literal and figurative understandings of the terms. Secondly, in view of the over-arching literary focus of this thesis, representations of borders and frontiers within postmodern literature are discussed. Thirdly, the disciplinary field of politics and literature and, subsequently, the political novel are examined. Then, mindful of the thematic tack of Rushdie's 2001 novel 'Fury', the real and imagined borders and frontiers governing processes of identity are explored primarily within the context of Rushdie's representation, within 'Fury', of the individual and the conflicting socioeconomic ethos of Western liberal democracies. Finally, the borders and frontiers of contemporary culture and celebrity are tested via perspectives which, in the first instance, are drawn from Rushdie's texts 'Fury' and 'Step Across This Line'. This thesis finds that Rushdie's description of his literary project, at least in terms of the two above-cited texts, is accurate. His writing is deeply characterised by a consciousness of borders and frontiers; however, rather than seeking to offer a clearly delineated, static or consistent account of them, his project is executed in a complex and sophisticated manner that presents a meaningful and distinctly human challenge to traditional notions of the nature and permeability of real and imagined borders and frontiers

    Analysis of circadian pattern reveals tissue-specific alternative transcription in leptin signaling pathway

    *Background*
It has been previously reported that most mammalian genes display a circadian oscillation in their baseline expression. Consequently, the phase and amplitude of each component of a signal transduction cascade has downstream consequences. 

*Results*
We report our analysis of alternative transcripts in the leptin signaling pathway which is responsible for the systemic regulation of macronutrient storage and energy balance. We focused on the circadian expression pattern of a critical component of the leptin signaling system, suppressor of cytokine signaling 3 (SOCS3). On an Affymetrix GeneChip 430A2 microarray, this gene is represented by three probe sets targeting different regions within the 3’ end of the last exon. We demonstrate that in murine brown adipose tissue two downstream 3’ probe sets experience circadian baseline oscillation in counter-phase to the upstream probe set. Such differences in expression patterns are a telltale sign of alternative splicing within the last exon of SOCS3. In contrast, all three probe sets oscillated in a common phase in murine liver and white adipose tissue. This suggests that the regulation of SOCS3 expression in brown fat is tissue specific. Another component of the signaling pathway, Janus kinase (JAK), is directly regulated by SOCS and has alternative transcript probe sets oscillating in counter-phase in a white adipose tissue specific manner.
 
*Conclusion*
We hypothesize that differential oscillation of alternative transcripts may provide a mechanism to maintain steady levels of expression in spite of circadian baseline variation

    Dose-finding study of a 90-day contraceptive vaginal ring releasing estradiol and segesterone acetate.

    ObjectiveTo evaluate serum estradiol (E2) concentrations during use of 90-day contraceptive vaginal rings releasing E2 75, 100, or 200 mcg/day and segesterone acetate (SA) 200 mcg/day to identify a dose that avoids hypoestrogenism.Study designWe conducted a multicenter dose-finding study in healthy, reproductive-aged women with regular cycles with sequential enrollment to increasing E2 dose groups. We evaluated serum E2 concentrations twice weekly for the primary outcome of median E2 concentrations throughout initial 30-day use (target ≥40 pg/mL). In an optional 2-cycle extension substudy, we randomized participants to 2- or 4-day ring-free intervals per 30-day cycle to evaluate bleeding and spotting based on daily diary information.ResultsSixty-five participants enrolled in E2 75 (n = 22), 100 (n = 21), and 200 (n = 22) mcg/day groups; 35 participated in the substudy. Median serum E2 concentrations in 75 and 100 mcg/day groups were <40 pg/mL. In the 200 mcg/day group, median E2 concentrations peaked on days 4-5 of CVR use at 194 pg/mL (range 114-312 pg/mL) and remained >40 pg/mL throughout 30 days; E2 concentrations were 37 pg/mL (range 28-62 pg/mL) on days 88-90 (n = 11). Among the E2 200 mcg/day substudy participants, all had withdrawal bleeding following ring removal. The 2-day ring-free interval group reported zero median unscheduled bleeding and two (range 0-16) and three (range 0-19) unscheduled spotting days in extension cycles 1 and 2, respectively. The 4-day ring-free interval group reported zero median unscheduled bleeding or spotting days.ConclusionsEstradiol concentrations with rings releasing E2 200 mcg/day and SA 200 mcg/day avoid hypoestrogenism over 30-day use.ImplicationsA 90-day contraceptive vaginal ring releasing estradiol 200 mcg/day and segesterone acetate 200 mcg/day achieves estradiol concentrations that should avoid hypoestrogenism and effectively suppresses ovulation

    Increased Drp1-mediated mitochondrial fission promotes proliferation and collagen production by right ventricular fibroblasts in experimental pulmonary arterial hypertension

    Introduction: Right ventricular (RV) fibrosis contributes to RV failure in pulmonary arterial hypertension (PAH). The mechanisms underlying RV fibrosis in PAH and the role of RV fibroblasts (RVfib) are unknown. Activation of the mitochondrial fission mediator dynamin-related protein 1 (Drp1) contributes to dysfunction of RV myocytes in PAH through interaction with its binding partner, fission protein 1 (Fis1). However, the role of mitochondrial fission in RVfib and RV fibrosis in PAH is unknown. Objective: We hypothesize that mitochondrial fission is increased in RVfib of rats with monocrotaline (MCT)-induced PAH. We evaluated the contribution of Drp1 and Drp1–Fis1 interaction to RVfib proliferation and collagen production in culture and to RV fibrosis in vivo. Methods: Vimentin (+) RVfib were enzymatically isolated and cultured from the RVs of male Sprague–Dawley rats that received MCT (60 mg/kg) or saline. Mitochondrial morphology, proliferation, collagen production, and expression of Drp1, Drp1 binding partners and mitochondrial fusion mediators were measured. The Drp1 inhibitor mitochondrial division inhibitor 1 (Mdivi-1), P110, a competitive peptide inhibitor of Drp1–Fis1 interaction, and siRNA targeting Drp1 were assessed. Subsequently, prevention and regression studies tested the antifibrotic effects of P110 (0.5 mg/kg) in vivo. At week 4 post MCT, echocardiography and right heart catheterization were performed. The RV was stained for collagen. Results: Mitochondrial fragmentation, proliferation rates and collagen production were increased in MCT-RVfib versus control-RVfib. MCT-RVfib had increased expression of activated Drp1 protein and a trend to decreased mitofusin-2 expression. Mdivi-1 and P110 inhibited mitochondrial fission, proliferation and collagen III expression in MCT-RVfib. However, P110 was only effective at high doses (1 mM). siDrp1 also reduced fission in MCT-RVfib. Despite promising results in cell therapy, in vivo therapy with P110 failed to prevent or regress RV fibrosis in MCT rats, perhaps due to failure to achieve adequate P110 levels or to the greater importance of interaction of Drp1 with other binding partners. Conclusion: PAH RVfib have increased Drp1-mediated mitochondrial fission. Inhibiting Drp1 prevents mitochondrial fission and reduces RVfib proliferation and collagen production. This is the first description of disordered mitochondrial dynamics in RVfib and suggests that Drp1 is a potential new antifibrotic target

    Weapons Make the Man (Larger): Formidability Is Represented as Size and Strength in Humans

    In order to determine how to act in situations of potential agonistic conflict, individuals must assess multiple features of a prospective foe that contribute to the foe's resource-holding potential, or formidability. Across diverse species, physical size and strength are key determinants of formidability, and the same is often true for humans. However, in many species, formidability is also influenced by other factors, such as sex, coalitional size, and, in humans, access to weaponry. Decision-making involving assessments of multiple features is enhanced by the use of a single summary variable that encapsulates the contributions of these features. Given both a) the phylogenetic antiquity of the importance of size and strength as determinants of formidability, and b) redundant experiences during development that underscore the contributions of size and strength to formidability, we hypothesize that size and strength constitute the conceptual dimensions of a representation used to summarize multiple diverse determinants of a prospective foe's formidability. Here, we test this hypothesis in humans by examining the effects of a potential foe's access to weaponry on estimations of that individual's size and strength. We demonstrate that knowing that an individual possesses a gun or a large kitchen knife leads observers to conceptualize him as taller, and generally larger and more muscular, than individuals who possess only tools or similarly mundane objects. We also document that such patterns are not explicable in terms of any actual correlation between gun ownership and physical size, nor can they be explained in terms of cultural schemas or other background knowledge linking particular objects to individuals of particular size and strength. These findings pave the way for a fuller understanding of the evolution of the cognitive systems whereby humans – and likely many other social vertebrates – navigate social hierarchies
